Supplements like niacinamide – a form of vitamin B3 – can help to boost NAD+ levels.

NAD+ boosting supplements are currently under investigation in the treatment of chronic diseases like obesity and type II diabetes.

And researchers have just discovered that time-of-day is important in regards to boosting NAD+ levels.

This means that those of us who take regular supplements of NAD+ boosters like niacinamide should take these early in the day.

The animal experiments were performed at the Instituto de Investigaciones Biomédicas, Universidad Nacional Autónoma de México. The paper was published as a preprint in bioRxiv.

“The circadian clock is a time-tracking endogenous system which anticipates and coordinates adaptation to daily environmental fluctuations.”

There are numerous mechanisms inside our bodies which tell us what time it is.

Light, food, temperature and other inputs all affect these clock settings.

Misalignment of this internal clock plays a large role in the development of chronic diseases…

Obesity and cancer are well known to be influenced by an out-of-time internal clock.

“Circadian misalignment leads to obesity, which is accompanied by reduced levels of the clock-controlled metabolite NAD+.”

We are learning that the central molecule in cellular energy production (NAD+) is tied to this internal clock.

So, researchers reasonably wondered whether this interaction could have knock-on effects for chronic disease.

“Increasing NAD+ levels is emerging as a therapy for diet-induced obesity and type 2 diabetes. The impact of daily fluctuations of NAD+ on these therapies remains unknown.

The researchers carried out experiments on mice to understand the relationship between our internal clocks and NAD+ influence on disease.

“Here, we demonstrate that time-of-day determines the efficacy of NAD+ as a therapy for diet-induced metabolic disease in mice.”

They discovered that boosting NAD+ at the start of the active phase helps with metabolic disease.

Mice become active when the sun goes down – but humans operate on a different time phase.

For us, the start of the active phase is in the morning.

“Restoring regular NAD+ oscillations at the onset of the active phase ameliorates metabolic markers of disease such as body weight and glucose and insulin tolerance, and restores hepatic gene expression related to inflammatory response and lipid metabolism.”

The researchers discovered that increasing NAD+ later in the waking day actually causes metabolic damage.

“However, the same treatment designed to increase NAD+ at the onset of the rest phase severely compromises these beneficial responses.”

Much of what was going on was occurring inside the liver.

The researchers discovered that boosting NAD+ at the wrong time of day upset sensing proteins – something which leads to a deranged metabolism.

“Notably, hepatic nutrient-sensing mTOR, AMPK or AKT signaling, became rhythmic specifically in obese mice treated just before the active phase.”

This research shows that taking supplements like niacinamide or other things which boost NAD+ should be done in the morning.

This knowledge could be the missing link for many people who may have been boosting their NAD+ levels later in the day and not getting the expected results.

“These findings demonstrate that the time of day determines the beneficial effects of NAD+-based therapies and pave the way for the basic strategy of a chronobiology-based therapeutic approach.”
