The cell experiments were performed at Missouri State University. This paper was published in Allergy.

I’ve written before about how increasing levels of carbon dioxide in the nasal cavity helps with symptoms of hay fever.

It does this by calming those cells in the nose.

“Intranasal non-inhaled delivery of carbon dioxide is efficacious in the symptomatic treatment of seasonal allergic rhinitis.”

The researchers were interested in what was going on a fundamental level.

They wanted to know if carbon dioxide could prevent mast cell degranulation.

Mast cells are immune cells which can release lots of histamine and other problematic substances.

High levels of histamine are responsible for the symptoms of hay fever and many of the symptoms of irritable bowel syndrome.

“The goal of this study was to determine whether and how 100% CO₂ inhibits mast cell degranulation, thereby possibly contributing to the reduction of symptoms in seasonal allergic rhinitis.”

The researchers carried out experiments on peritoneal cells.

These are cells taken from the membrane that lines the inside of your abdomen and pelvis…

Cells which can become irritated and release lots of histamine causing a wide array of symptoms.

“Peritoneal mast cells isolated from rats were used to determine whether CO₂ treatment could block mast cell degranulation and histamine release in response to 48/80.”

48/80 is a synthetic chemical which triggers the release of histamine from mast cells.

It imitates the effects of many naturally occurring irritants and allergens.

48/80 caused the mast cells to degranulate – releasing massive amounts of histamine.

“Treatment with 48/80 caused >90% of mast cells to degranulate causing a significant increase in histamine release.”

But when the cells had access to lots of CO2, degranulation was prevented.

Keeping histamine inside the mast cells where it doesn’t do any harm.

“Significantly, the stimulatory effect of 48/80 on fluorescent intensity and histamine levels was greatly inhibited (>95%) to near control levels by pretreatment with 100% CO₂.”

The researchers discovered that these effects were related to the movement of calcium into the cell caused by the irritant.

Something which was prevented by carbon dioxide.

“Treatment with 48/80 also caused a robust transient increase in intracellular calcium, whereas pretreatment with CO₂ repressed the increase in calcium (>70%) in response to 48/80.”

Carbon dioxide helps with hay fever and other symptoms of excess histamine by stabilizing the cell, preventing the erratic flow of calcium, and thereby reducing the release of histamine.

“Results from this study provide the first evidence of a unique regulatory mechanism by which CO₂ inhibits mast cell degranulation and histamine release by repressing stimulated increases in intracellular calcium.”

Carbon dioxide is now used as a topical treatment inside the nasal cavity for a fever.

Note that it’s not inhaled – inhaling high concentrations of CO2 can be lethal.

The treatment involves letting it absorb into the skin and the nasal cavity…

…saturating those cells with CO2, stabilizing them and thereby preventing the release of histamine.

“Thus, our data provide a plausible explanation for the reported therapeutic benefit of non-inhaled intranasal delivery of 100% CO₂ to treat allergic rhinitis.”
