The Alzheimer’s Association reports that the incidence of Alzheimer’s is increasing and will continue to do so because of our aging population.


Between the ages of 65 and 85, the chance of developing dementia goes from 1 in 25 to 1 in 3.

Advances in genetic medicine have had little success with dementia.

The outlook seems bleak.

However, a recent finding shows that some old treatments can not only stop the signs and symptoms of dementia but reverse them.

This study, published in the journal Nature, investigated the role of tau fibrils in neurodegenerative disorders.

In conditions like Alzheimer’s and temporofrontal dementia, tau fibrils accumulate in the brain.

As these tau structures increase, a person’s mental faculties decrease.

Recently, scientists have identified a soluble subtype of these structures… it seems to be the one most involved in causing structural and cognitive damage.

This paper identified one of the key chemical changes in tau structure that leads to the toxic soluble tau type found in dementias like Alzheimer’s.

“Here we identify tau acetylation as an early change in Alzheimer’s brains.”

This important discovery allowed the scientists to develop a mechanism to prevent these damaging changes in the brain.

Here’s what they did:

First, they created a mutant mouse.

This mouse was designed to have increased acetylation.

That’s the process which had been identified as the cause of toxic tau formation.

As expected, the mutant mouse had altered tau formation and the cognitive deficits associated with dementia – confirming the mechanism in a live animal.

“The acetyl-mimicking mutant slows tau turnover and induces cognitive deficits.”

Previous work has identified the enzyme that causes acetylation of the toxic tau structures.

A number of drug-based inhibitors of this process were already known (salicylate and salsalate).

The scientists then tested these inhibitors.

“Tau acetylation is inhibited by salsalate and salicylate, which enhance tau turnover and reduce tau levels.”

The treatments worked as expected. The results were astonishing.

Both of these treatments reduced the amount of tau buildup!

The administration of salsalate lowered levels of tau in the dementia-stricken mice.

It reversed physical indicators of dementia in the brain.

“Administration of salsalate after disease onset lowered levels of total tau.”

Not only did the treatment reverse the damage to the brain, but cognitive performance was improved.

Salsalate restored lost memory capability.

“Salsalate after disease onset rescued tau-induced memory deficits.”

One of the key features of Alzheimer’s disease is a loss of brain mass in the hippocampus region of the brain.

This atrophy is thought to be due to a loss of critical neurons as a result of the formation of toxic fibrils like tau.

The study found that the drug prevented loss of brain mass in this region.

“Salsalate after disease onset prevented hippocampal atrophy.”

The study concludes that targeting tau acetylation in this manner could be a valuable tool in fighting the degenerative brain disorders that are caused by these tangled tau fibrils.

“Targeting tau acetylation could be a new therapeutic strategy against human tauopathies.”

You may have recognized the names of one or more of those treatments.

Salicylate and salsalate.

Both are potent anti-inflammatories which have been known for over a hundred years.

Salicylate or salicylic acid is a primary plant hormone found widely in nature.

There are very high amounts of it in the bark of the willow tree.

Willow has been used medicinally for thousands of years because of its salicylic acid content.

Both of these drugs are very closely related to aspirin.

Aspirin’s chemical name is acetylsalicylic acid – it is an acetyl group attached to salicylate.

In fact, a lot of aspirin’s effectiveness is due to salicylic acid.

The body breaks aspirin down into an acetyl group and salicylic acid.

The only difference is that you would need to take slightly more aspirin to get the same dose of salicylic acid (salicylate).

So yet again we see that one of the oldest drugs could be re-purposed as a cure for a modern epidemic.

Aspirin has been out of patent for a long time.

This may be why it’s not on the front page of the news as often as it should be.
