How DHT protects men

Graves’ disease is a common thyroid condition.

It is an autoimmune disease like the more well-known Hashimoto’s…

But it has opposing effects on thyroid hormones compared with that diagnosis.

Graves’ disease causes abnormally high levels of thyroid hormones. 

It can also lead to other symptoms affecting the skin and eyes.

Like many diseases involving autoimmunity, it does not affect men at the same rate as it does women — this is partly due to hormones…

However…

It has been known for some time that estrogen can increase the risk of autoimmune disease.

But male hormones like DHT have calming effects on the immune system

So researchers wondered whether increasing DHT could have the opposite effect of estrogen – reducing the risk of Graves’ disease.

Their animal experiments showed that this may just be the case.

The rodent experiments were carried out at The First Affiliated Hospital of Xi’an Jiaotong University Health Science Center in Xian China. The paper was published in the journal Thyroid.

“Graves’ disease (GD) is a common organ-specific autoimmune disease characterized by hyperthyroidism that has significant sex differences in prevalence and clinical expressions.”

Graves is a disease of the immune system which has its effects on the thyroid gland.

“Abnormal cytokine production and T cell activation may result in various manifestations of GD.”

But androgenic hormones like DHT can have calming effects on the immune system.

Some research has shown that they could even protect against autoimmune diseases similar to Graves’.

“Studies have shown that androgen treatment can provide protection against autoimmune diseases. But as yet, the effects of androgen treatment on GD are still unknown.”

So these researchers decided to find out if there were any effects on the risks of Graves’ disease related to levels of DHT.

“This study investigated whether a potent bioactive androgen, DHT, could be of benefit in a mouse model of GD.”

They decided to give some animals DHT before provoking an immune response that could trigger Graves’ disease.

They wanted to see if and how DHT could alter Graves’ disease risk.

“We investigated (i) whether DHT pretreatment inhibits autoimmune responses, and (ii) the mechanism of immune protection of DHT in GD.”

The research was carried out on female mice which were given a modified virus designed to trigger autoimmunity, which causes Graves’ disease.

“Female mice were immunized three times with an adenovirus expressing the human thyrotropin receptor.”

Three groups of mice were given increasing doses of DHT.

Another group was not given any supplemental hormones but still exposed to the viral trigger.

“Three doses (1.5, 5, and 15 mg) of DHT or a matching placebo were implanted a week before the first immunization.”

Four weeks after all parts of the experiment were completed, the researcher sacrificed animals after taking blood samples.

The primary problem with Graves’ disease is high levels of thyroid hormones.

The thyroid hormones were much lower in the mice which had been pretreated with DHT.

“After DHT treatment, thyroid hormones were dramatically reduced compared with placebo.”

The researchers also found far less signs of autoimmunity in the animals.

Interferon Y and interleukin-2 are elevated in Graves’ disease – but this was blunted by pretreatment with DHT.

“In addition, a remarkable reduction in interferon-γ and interleukin-2 production was observed in DHT-pretreated mice.”

Other tests show that DHT directly down-regulates some of the most important aspects of the overactive immune system.

“DHT can alleviate the severity of GD by downregulating pro-autoimmune T helper 1 cells in female mice.”

And finally, the researchers found that the higher doses of DHT were more effective.

“The protective influence was more noticeable with 5 mg and 15 mg doses of DHT.”

The study could provide a new line of treatment for men with Graves’ disease if they have low DHT.

But DHT also works similar to other hormones which could help other men and even women to control their autoimmune flares.
